Acetyl-l-carnitine is a stabilized form of the amino acid metabolite L Carnitine and is utilized in the same manner in the body. acetyl-l-carnitine plays an important role in protecting and supporting brain functions.
Benefit of acetyl-l-carnitine
The mind boosting effect of acetyl-l-carnitine is often noticed within a few hours, or even within an hour. Most people report feeling mentally sharper, having more focus and being more alert. Some find a mild mood enhancement. acetyl-l-carnitine may be used as an overall mind booster. The typical dosage is 250 mg once a day or a few times a week. Another option is 300 mg. If these dosages are not enough to make you more mentally alert, consider, 500 mg.

Modulatory effect of acetyl-L-carnitine on amyloid
precursor protein metabolism in hippocampal neurons.
Eur J Pharmacol. 2008. Epis R, Marcello E, Gardoni F, Longhi A, Calvani M, Iannuccelli M, Cattabeni F, Canonico PL, Di Luca M. Department of Pharmacological Sciences and Center of Excellence on Neurodegenerative Diseases, University of Milan, Milan, Italy.
Alzheimer Disease is the most common chronic neurodegenerative disorder associated with aging. Nevertheless, its pharmacological therapy is still an unresolved issue. In double-blind controlled studies, acetyl-L-carnitine demonstrated beneficial effects on Alzheimer's disease. However, the mechanisms behind its neuroprotective ability remain to be fully established. In this study, the effect of acetyl-L-carnitine on amyloid precursor protein (APP) metabolism was investigated by in vitro models, both in a neuroblastoma cell line and in primary hippocampal cultures. We found that acetyl-L-carnitine treatment stimulates alpha-secretase activity and physiological APP metabolism. In particular, this brain product favors the delivery of ADAM10 (a disintegrin and metalloproteinase 10, the most accredited candidate for alpha-secretase) to the post-synaptic compartment, and consequently positively modulates its enzymatic activity towards APP. Our findings suggest that the benefits of acetyl-L-carnitine reported in previous clinical studies are underscored by the specific biological mechanism of this compound on APP metabolism. In fact, acetyl-L-carnitine can directly influence the primary event in Alzheimer's disease pathogenesis, i.e. the Amyloid beta cascade, promoting alpha-secretase activity and directly affecting the release of the non amyloidogenic metabolite.

A double-blind, randomised, controlled clinical trial of acetyl-L-carnitine vs. amisulpride in the treatment of dysthymia.
Eur Neuropsychopharmacol. 2006. Department of Psychiatry, School of Medicine, Vita-Salute University San Raffaele, Milan, Italy.
Researchers evaluated the effect of acetyl-L-carnitine ( ALCAR ) vs. amisulpride measured by total Hamilton Depression Rating Scale score in patients with pure dysthymia. Two hundred and four patients were randomised and treated with ALCAR 500 mg twice a day or amisulpride 50 mg u.i.d. in a double-blind study, for 12 weeks. A solid improvement was observed in both treatment groups throughout the study. The results did not disclose statistically significant differences between treatments. The greater tolerability of ALCAR is of clinical relevance considering the chronicity of dysthymia, which often requires prolonged treatment.
ADD or ADHD treatment
ADHD is not easy to treat, supplements that may be helpful in reducing symptoms have not been well tested, but ALC may offer some benefit.
Evaluation of lenticular antioxidant and redox system components in the lenses of acetyl-L-carnitine treatment in BSO-induced glutathione deprivation.
Mol Vis. 2009; Elanchezhian R, Sakthivel M, Isai M, Geraldine P, Thomas PA. Department of Animal Science, School of Life Sciences, Bharathidasan University, Tamil Nadu, India.
To investigate whether acetyl-L-carnitine (ALCAR) retards L-buthionine-(S,R)-sulfoximine (BSO)-induced cataractogenesis in Wistar rat pups. ALCAR prevents cataractogenesis in the BSO-induced cataract model, possibly by inhibiting depleting antioxidant enzyme and redox system components and inhibiting lipid peroxidation.
I have had on-going heavy duty depression since I was 18 yrs old. I don't have those ups and downs that I've heard so much about. Just an ongoing depression at different levels of "heaviness" throughout the day, day after day. At its heaviest point it's like my mind and body wants to shut off to the outside world. Vast majority of the time it's like my mind and body simply cannot produce energy. Here's the regimen I'm following that helps: In the mornings I'm taking 500 mg of Acetyl L-Carnitine with 2 cups of warm tea and 500 mg of l-carnitine l-tartrate along with 5-HTP 50 mg in the evening. It sure seems that I have nutritional needs that ought to be met daily.
Acetyl l-carnitine for
Researchers at the Polytechnic University of Marche, in Italy had an inkling that supplements could make a difference. They evaluated sixty infertile men between the ages of 20 and 40 who had poor sperm motility or movement. They gave these men the nutrients carnitine, acetyl l-carnitine, or a combination. The results showed that the combination of carnitine and acetyl l-carnitine had the best outcome with improvement noted in sperm motility and velocity. The antioxidant capacity of the sperm fluid also increased.
It would seem reasonable for men who have undergone a medical evaluation for infertility and have not had success with the standard medical approach to try a combination of these two nutrients, particularly if analysis shows their sperm do not swim well or are slow movers. The ideal dosage is difficult to know, but a cautious approach would be to alternate taking 200 to 300 mg of acetyl l-carnitine one day and 200 to 300 mg of carnitine the following day. The dosage can be increased after a month or two if the results are not adequate. Higher doses could lead to overstimulation, restlessness, and insomnia.
Acetyl l carnitine and alpha
lipoic acid for Parkinson's disease
Combined R-alpha-lipoic acid and acetyl-L-carnitine exerts efficient preventative effects in a cellular model of Parkinson's disease.
J Cell Mol Med. 2008. Institute for Nutritional Science, Shanghai Institutes of Biological Sciences, Chinese Academy of Sciences, Shanghai, China.
Mitochondrial dysfunction and oxidative damage are highly involved in the pathogenesis of Parkinson's disease. Some mitochondrial antioxidants / nutrients that can improve mitochondrial function and/or attenuate oxidative damage have been implicated in PD therapy. However, few studies have evaluated the preventative effects of a combination of mitochondrial antioxidants/nutrients against Parkinson's disease, and even fewer have sought to optimize the doses of the combined agents. The present study examined the preventative effects of two mitochondrial antioxidant/nutrients, R-alpha-lipoic acid (LA) and acetyl-L-carnitine, in a chronic rotenone-induced cellular model of PD. We demonstrated that 4-week pretreatment with R-alpha-lipoic acid and/or acetyl-L-carnitine effectively protected SK-N-MC human neuroblastoma cells against rotenone-induced mitochondrial dysfunction, oxidative damage, and accumulation of alpha-synuclein and ubiquitin. Most notably, we found that when combined, R-alpha-lipoic acid and acetyl-L-carnitine worked at 100 to 1000 fold lower concentrations than they did individually. We also found that pretreatment with combined R-alpha-lipoic acid and acetyl-L-carnitine increased mitochondrial biogenesis and decreased production of reactive oxygen species through the upregulation of the peroxisome proliferator-activated receptor-gamma coactivator 1alpha as a possible underlying mechanism. This study provides important evidence that combining mitochondrial antioxidant / nutrients at optimal doses might be an effective and safe prevention strategy for PD.
Side effects of acetyl-l-carnitine include overstimulation at dosages greater than 500 mg. Nausea is another acetyl-l-carnitine side effect. In a few people, headache may be a cause of from a high dose.
How often to take an acetyl-carnitine
Acetyl-l-Carnitine 300 mg is a potent mind booster and antioxidant. Most people find one capsule is the right dose while some people may find the occasional use of 2 capsules helpful. Acetylcarnitine increases alertness and focus. It would be best not to take a capsule with other brain boosting supplements due to possible overstimulation. One option is to alternate the use of acetylcarnitine one day with Mind Power Rx the next day, or alternate with other brain supplements such as DMAE, choline, ginkgo, etc. Doses above 500 mg may, in some people, lead to overstimulation, insomnia, and nausea. For long term use one capsule 3 times a week with a week off each month is a good option. Acetylcarnitne can be taken with small doses of other supplements such as vitamins E and C, and it can be used together with fish oils. Many products on the market have acetylcarnitine together with alpha lipoic acid and are promoted for antiaging purposes. Be careful using high amounts of these combinations since they can keep you awake at night, and shallow sleep is certainly not helpful for longevity purposes. We would not recommend taking more than 10 to 30 mg of R lipoic acid or 20 to 60 mg of regular alpha lipoic acid when combined with 300 mg of acetylcarnitine.

The association of acetyl-l-carnitine and nicotinamide remits the experimental diabetes in mice by multiple low-dose streptozotocin.
Pancreas. 2006. Centro de Investigaciones Endocrinologicas (CEDIE-CONICET), Htal. de Ninos R. Gutierrez, Buenos Aires, Argentina.
We studied the effect of acetyl-l-carnitine plus nicotinamide on murine diabetes mellitus induced by multiple low doses of streptozotocin. Male C57BL/6J inbred mice were injected intraperitoneally with citrate buffer or streptozotocin (40 mg/kg) for 5 consecutive days, followed by injections of saline solution or acetyl-l-carnitine plus nicotinamide (50 + 25 mg/kg) from days 6 to 110. Four groups were studied: normal control mice (C), treated normal control mice (TC), diabetic mice (D), and treated diabetic mice (TD). The treatment with acetyl-l-carnitine plus nicotinamide induced remission of autoimmune type 1 diabetes in mice produced by multiple low doses of streptozotocin.

There is no human evidence that the combination of these two nutrients is better than either alone. Even if the combination is preferable, we have no idea of the ideal dosage, and it could be vastly different between different people. Both acetyl-l-carnitine and alpha lipoic acid can be taken with food to reduce nausea, but if one wishes to feel the mental stimulating effects more clearly, they can be taken on an empty stomach.
Q. Does acetyl-l-carnitine help
